The agreement tasks Amberstone with generating preclinical candidates against two targets selected by Henlius. Following this initial phase, Henlius will assume control over global research, development, manufacturing, and commercialization. Financial terms include an upfront payment and research funding for Amberstone, with potential milestone payments reaching up to $440 million per target, in addition to tiered royalties.
Traditional T-cell engagers often suffer from severe side effects, including cytokine release syndrome and off-tumor toxicity. Amberstone’s T-MATE technology addresses these hurdles via reversible pH gating. This mechanism ensures the therapy remains largely inactive in the bloodstream and healthy tissues, activating only within the acidic microenvironment characteristic of solid tumors. Amberstone expects its own lead asset developed through this platform to reach clinical trials by the first half of 2027.
