Jazz Pharmaceuticals Phase 3 LAGOON Trial Fails Primary Endpoint

Jazz Pharmaceuticals announced that its Phase 3 LAGOON trial failed to meet its primary endpoint of overall survival for Zepzelca in patients with relapsed metastatic small cell lung cancer. The study evaluated the drug as both a monotherapy and in combination with irinotecan against standard investigator-selected treatments.

The trial, conducted by partner PharmaMar, involved 724 patients across more than 200 global sites. Despite the lack of efficacy in this specific second-line setting, the company confirmed that no new safety signals emerged. The results showed that the drug's safety profile remained consistent with previous data, though the primary goal of extending survival compared to current options was not achieved.

Rob Iannone, executive vice president of research and development at Jazz Pharmaceuticals, stated that the company continues to view Zepzelca as a critical treatment for small cell lung cancer, specifically in the first-line maintenance setting. He noted that the LAGOON results are distinct from the successful IMforte trial, which previously supported the drug's U.S. approval. Jazz has shared the findings with the FDA and plans to discuss the next steps regarding post-marketing requirements for the second-line indication. The company emphasized that these trial outcomes do not impact its 2026 financial guidance.
